In creating a new biomarker for Chlamydomonas, it was determined that arginine biosynthesis occurs in the chloroplast of this green alga, as transforming the chloroplast genome of the arg9-2 nuclear mutant with a wildtype ARG9 gene restored arginine prototrophy.
LTO1 was identified by sequence homology to VKOR (vitamin K epoxide reductase). Using the LTO1 gene from Arabidopsis thaliana, it was shown that Lto1 functionally complemented an E. coli mutant deficient in periplasmic disulfide bond formation and also reduced Photosystem II subunit PsbO in vitro.
Finally, I worked with C. reinhardtii as a prototype to unravel the system II maturation pathway of c-type cytochromes in the chloroplast using forward genetics. The c-type cytochromes are a class of heme containing electron shuttles in which the heme co-factor is covalently attached via thioether bonds at a heme binding site defined by a CXXCH motif. They are crucial to basic cellular functions such as photosynthesis and respiration, and yet the molecular details of their assembly are not clearly understood.
